The outline of Tetragenococcus halophilus NBRC 12172 proteome
We have analyzed the proteome of halotolerant lactic acid bacterium, Tetragenococcus halophilus NBRC 12172, cultured with added salt concentration of 0% and 10% by using the following methods.
- (1) Soluble fraction: Two-dimensional PAGE followed by MALDI-TOFMS
- (2) Soluble fraction: One-dimensional SDS-PAGE in combination with two-dimensional LC-MS/MS
- (3) Soluble fraction: Whole shotgun proteomics
- (4) Membrane enriched fraction (treated by lysozyme and ultracentrifuged): One-dimensional SDS-PAGE in combination with two-dimensional LC-MS/MS
- (5) Membrane enriched fraction (disrupted on dry ice and ultracentrifuged): on-column digestion followed by LC/MS/MS
- (6) Membrane enriched fraction (disrupted on dry ice): on-column digestion followed by LC/MS/MS
N-terminal amino acid sequencing by the protein sequencer were also performed.
The result of proteome analysis
Tetragenococcus halophilus NBRC 12172 cultured under 0% salt condition was in more stressed condition rather than in 10% condition. It indicated that this strain acquires energy using high salt condition environment. In addition, it was suggested that the salt resistance was related to uptake of osmoregulation substance and membrane glycoprotein.
